Transportation in the Bay Area falls somewhere between New York and LA: you can get around without a car, but it's not exactly quickest or most pleasant way to do things. On the other hand, traffic on 101 can be a nightmare during rush hour, or at six PM on a Saturday, or even at one AM.
That being said, if you're planning on leaving San Francisco, especially to go to Napa, you will want a car. All major rental agencies rent out of SFO, OAK, and SJC airports, and have very reasonable rates, especially on the weekend. A word of warning, though: hotel parking in San Francisco is usually expensive.
